Hybrids combining desirable traits from divergent parents are the main objective of some Brachiaria (Syn. Urochloa P. Beauv.) breeding programs. There is great interest in the development of apomictic hybrid cultivars that combine desirable genes such as resistance to spittlebugs, high nutritive value, and tolerance to acid soils. Microsporogenesis of six apomictic progenies resulting from a tetraploid (2n = 4x = 36) cross between B. ruziziensis × B. brizantha was evaluated under light microscopy. Genetic recombination, ensured by multivalent chromosome association and crossing-over at prophase occurred in low frequency among progenies, and in one, recombination was almost nonexistent. The percentage of meiocytes with meiotic abnormalities among progenies ranged from 16.6 % to 85.6 %. Besides an observed irregular chromosome segregation typical of polyploid hybrids in these five progenies, putative meiotic mutations characterized as desynapsis and divergent spindle organization occurred in three progenies. These anomalies caused frequent fractionation of the genome into several microspores of different sizes. In Brachiaria, new cultivars must be apomictic to fix the genotype. However, Brachiaria is a pseudogamous apomict, and viable gametes are necessary to produce viable seeds. Considering meiotic behavior, only two progenies are promising for advancement in the breeding program.

Panicum maximum Jacq. é uma das gramíneas mais cultivadas no mundo. O programa de hibridização intraespecífica, desenvolvido pela Embrapa Gado de Corte objetiva combinar diversas características agronômicas em uma única planta com alta produção de sementes. Quatro híbridos intraespecíficos, originados a partir do cruzamento entre fêmeas sexuais tetraploidizadas artificialmente com plantas apomíticas, incluindo os respectivos genitores, foram citologicamente avaliados em seu processo meiótico por meio de metodologia convencional. Híbridos e genitores apresentaram alta estabilidade meiótica, e a percentagem de anormalidades meióticas entre eles variou de 6,7 a 14,2%. As anormalidades encontradas foram segregação irregular de cromossomos, aderências e ausência de citocinese, mas esta última não comprometeu a viabilidade do pólen. Configurações cromossômicas em multivalentes na diacinese, que variam de um a quatro quadrivalentes, sugerem que a recombinação e a introgressão de alguns genes pode ser esperada em híbridos. Com base na estabilidade meiótica observada, os híbridos poderão ser selecionados para alta produção de sementes, e os genitores podem permanecer no programa de melhoramento participando em outros cruzamentos intraespecíficos.

Panicum maximum Jacq. is one of the most cultivated grasses in the world. The intraspecific hybridization breeding program underway at Embrapa Beef Cattle aims at combining several agronomic characteristics in a single plant with high seed production. Four intraspecific hybrids originated from the cross between sexual tetraploid female plants with apomictic tetraploid male plants, including the genitors, were cytologically evaluated in the meiotic process throughout conventional methodology. Hybrids and genitors showed high meiotic stability. The percentage of abnormal cells among them ranged from 6.7 to 14.2%. Abnormalities recorded were irregular chromosome segregation, chromosome stickiness, and absence of the first cytokinesis, but the latter did not compromise pollen viability. Multivalent chromosome configurations at diakinesis, ranging from one to three quadrivalents, suggest that genetic recombination and introgression of some genes can be expected in the hybrids. Based on meiotic stability, the hybrids should be selected for high seed production, and the genitors could remain in the program participating in other intraspecific crosses.

O comportamento meiótico de quatro híbridos interespecíficos promissores de Brachiaria foi avaliado por metodologia citogenética convencional. Os genitores femininos eram dois acessos sexuais tetraploidizados artificialmente de B. ruziziensis (R41 e R44, 2n = 4χ = 36), os quais foram cruzados com um genσtipo tetraplσide natural apomítico agronomicamente superior de B. brizantha (B140 - BRA003395). Três deles (HBGC313, HBGC 315 e HBGC324) eram sexuais e um (HBGC325) apomítico. A análise de algumas células em diacinese revelou a presença de configurações cromossômicas multivalentes, sugerindo que recombinação genética e introgressão de alguns genes poderão estar presentes. Os quatro híbridos apresentaram freqüências e tipos variados de anormalidades meióticas. Anormalidades relacionadas à segregação irregular de cromossomos devido à poliploidia foram comuns entre os híbridos e caracterizadas pela presença de cromossomos em ascensão para os pólos e cromossomos retardatários, ambos levando à formação de micronúcleos em telófases e tétrades e, como conseqüência, gametas desbalanceados. Uma anormalidade genótipo-específica, relacionada à orientação do fuso (uma possível mutação fuso divergente) foi observada pela primeira vez em dois dos híbridos, HBGC313 e HBGC325. O híbrido sexual HBGC324 apresentou menor freqüência de anormalidades meióticas, podendo ser usado como genitor feminino em futuros cruzamentos no programa de melhoramento. A frequência e os tipos de anormalidades observadas podem afetar a fertilidade e a produção de sementes. Com base nos resultados, o híbrido HBGC324 é recomendado para o programa de melhoramento. Híbridos devem produzir grande quantidade de sementes viáveis, além de matéria seca com alto valor nutritivo para serem amplamente utilizados na produção de pastagens. Devido à pseudogamia, os híbridos apomíticos superiores necessitam de grãos de pólen viáveis para fertilizar o núcleo secundário do saco embrionário assegurando o desenvolvimento vigoroso do endosperma e a produção de sementes.

The meiotic behavior of four interspecific promising hybrids was evaluated by conventional cytological methods. The female genitors were two artificially tetraploidized sexual accessions of B. ruziziensis (R41 and R44, 2n = 4χ = 36), which were crossed to an agronomically superior natural tetraploid apomictic genotype of B. brizantha (B140 - BRA003395). Three of them (HBGC313, HBGC 315, and HBGC324) were sexual and one (HBGC325) apomictic. Analyses of some cells in diakinesis revealed multivalent chromosome configurations, suggesting that genetic recombination and introgression of some genes could be present. The four hybrids had different types of meiotic abnormalities at various frequencies. Abnormalities related to irregular chromosome segregation due to polyploidy were common among these hybrids, and characterized by precocious chromosome migration to the poles, laggard chromosomes, both generating micronuclei in telophases and tetrads and, as a consequence, unbalanced gametes. One abnormality genotype-specific, related to spindle orientation (a putative divergent spindle mutation), was recorded for the first time in two of the hybrids, HBGC313 and HBGC325. The sexual hybrid HBGC324 had the lower rate of abnormalities, and it could be used as a female genitor in future crosses in the breeding program. The abnormalities present in these hybrids may impact fertility and affect seed production. Based on the results, HBGC324 is the single hybrid recommended to the breeding program. Hybrids must produce a good amount of viable seeds, besides good overall dry matter production and nutritive value, in order to be widely utilized and adopted in production systems. Due to pseudogamy, the desirable superior apomictic hybrids need viable pollen grains to fertilize the secondary nucleus of the embryo sac and thus ensure normal and vigorous endosperm development and plenty of seed set.

Algumas espécies africanas de Brachiaria foram introduzidas nas Américas e tornaram-se importantes pastagens nos trópicos. Novas cultivares podem ser obtidas por seleção direta da variabilidade genética existente na coleção de germoplasma ou por hibridização interespecífica. Poliploidia é predominante no gênero Brachiaria e correlacionada com apomixia, o que complica a hibridização. O objetivo dos estudos citogenéticos na coleção de germoplasma de Brachiaria da Embrapa Gado de Corte é determinar o número de cromossomos e o comportamento meiótico dos acessos. A microsporogênese foi avaliada em dois acessos de Brachiaria bovonei (BO01 e BO05) e um acesso de B. subulifolia (SU01). BO01 é pentaplóide (2n = 5x = 45), BO05 é tetraplóide (2n = 4x = 36), e SU01 é hexaplóide (2n = 6x = 54), todos derivados de x = 9. Anormalidades meióticas típicas de poliplóides, caracterizadas por migração precoce de cromossomos para os pólos em metáfases, cromossomos retardatários em anáfases, e micronúcleos em telófases e tétrades foram observadas em alta freqüência em todos os acessos gerando gametas geneticamente desbalanceados. Ambos os acessos de B. bovonei apresentaram aderências cromossômicas. Os resultados são discutidos sob o ponto de vista dos objetivos do melhoramento genético.

Some African species of Brachiaria have been introduced into the Americas and became the most important forage for pastures in the tropics. New cultivars can be obtained either from direct selections from the natural existing variability in the germplasm collections or from interspecific hybridizations. Polyploidy is predominant in the genus Brachiaria and correlated with apomixis which complicates hybridization. The objective of cytological studies underway on the Brachiaria germplasm collection at Embrapa Beef Cattle is to determine the chromosome number and meiotic behavior of accessions. For the breeding of Brachiaria species, compatible sexual and apomictic accessions need to be identified. Microsporogenesis was evaluated in two accessions of Brachiaria bovonei (BO01 and BO05) and one accession of B. subulifolia (SU01). BO01 is pentaploid (2n = 5x = 45), BO05 is tetraploid (2n = 4x = 36), and SU01 is hexaploid (2n = 6x = 54), derived from x = 9. Meiotic abnormalities typical of polyploids, characterized by precocious chromosome migration to the poles in metaphases, laggard chromosomes in anaphases, and micronuclei in telophases and tetrads, were recorded in high frequency in all the accessions generating unbalanced gametes. Both accessions of B. bovonei presented chromosome stickiness. The results are discussed in the view of the Brachiaria breeding program objectives.

Os dois acessos de B. dura analisados (DU01 e DU02) são hexaplóides (2n = 6x = 54), derivados de x = 9. Anormalidades meióticas como migração precoce de cromossomos para os polos, cromossomos retardatários e micronúcleos foram observados em baixa frequência em ambos os acessos. A presença de poucas associações cromossômicas em diacinese e a estabilidade meiótica sugere que a hexaploidia provavelmente resultou de duplicação cromossômica. No acesso DU02 observou-se transferência de cromossomos (citomixia) entre meiócitos, envolvendo parte ou todo o genoma. As implicações destes resultados para o melhoramento de Brachiaria são discutidas.

The two accessions of B. dura analyzed (DU01 and DU02) are hexaploid (2n = 6x = 54), derived from x = 9. Meiotic abnormalities, such as precocious chromosome migration to the poles, laggards and micronuclei, were recorded in low frequency in both accessions. The few multivalent chromosome association at diakinesis and meiotic stability suggested that hexaploidy probably resulted from chromosome doubling. In DU02, chromosome transfer (cytomixis) among meiocytes, involving part or the entire genome was observed. The implication of these findings for the Brachiaria breeding is discussed.

A microsporogênese de um híbrido interespecífico entre um acesso sexual tetraploidizado artificialmente de Brachiaria ruziziensis (2n=4x=36) e um acesso apomítico tetraplóide natural de B. decumbens (2n=4x=36) foi analisada. Sincícios envolvendo um grande número de células foram encontrados em 15,40% dos meiócitos. A meiose progrediu normalmente nos sincícios durante a prófase I; em metáfase I, todavia, muitos núcleos fundiram-se, mostrando ainda aderências cromossômicas e inúmeros fragmentos. O processo meiótico foi interrompido na metáfase I, quando a cromatina formou núcleos picnóticos. Citocineses anormais fracionaram os sincícios em produtos meióticos anômalos que foram recobertos pela parede do grão de pólen. Meiócitos em leptóteno também foram observados durante todo o processo meiótico e grãos de pólen anormais com parede de pólen bem desenvolvida, mas contendo núcleos leptotênicos, foram observados em 9,18% dos grãos de pólen analisados. Os resultados sugerem que os meiócitos receberam o sinal para entrar em meiose, mas não receberam o sinal para prosseguir além do leptóteno. Apesar da ausência de processo meiótico completo, os meiócitos foram cobertos pela parede do grão de pólen. Estas anormalidades, combinadas com outras causadas pela poliploidia, resultaram em total esterilidade de pólen.

Microsporogenesis was analyzed in an interspecific hybrid between an artificially tetraploidized sexual accession of Brachiaria ruziziensis (2n=4x=36) and a natural apomictic tetraploid accession of B. decumbens. Syncytes involving a large number of cells were recorded in 15.4% of meiocytes. Meiosis progressed normally in syncytes during prophase I; in metaphase I, however, several nuclei were found fusioned, showing chromosome stickiness and several chromosome fragments. Meiosis was arrested in metaphase I and pycnotic nuclei and micronuclei were formed. Abnormal cytokinesis fractionated the syncyte into abnormal meiotic products that were covered by the pollen wall. Meiocytes in leptotene were recorded in all the slides prepared for both meiotic divisions, and abnormal "pollen grains" with well-developed pollen wall but containing leptotene nuclei were recorded in 9.18% of grains analyzed. These findings suggested that the meiocytes received the signal to enter meiosis but lacked the signal to proceed beyond leptotene. Despite the absence of the meiotic process, such cells were covered by pollen grain wall. Total pollen sterility resulted from these abnormalities combined with still others observed among meiocytes.

This paper reports a case of abnormal spindle orientation during microsporogenesis in an interspecific hybrid of the tropical grass Brachiaria. In the affected plant, prophase I was normal. In metaphase I, bivalents were regularly co-oriented but distantly positioned and spread over the equatorial plate. In anaphase I, chromosomes failed to converge into focused poles due to parallel spindle fibers. As a consequence, in telophase I, an elongated nucleus or several micronuclei were observed in each pole. In the second division, the behavior was the same, leading to polyads with several micronuclei. A total of 40% of meiotic products were affected. The use of this hybrid in production systems needing good-quality seeds is discussed.

Microsporogenesis was analyzed in an interspecific hybrid between an artificially tetraploidized sexual accession of Brachiaria ruziziensis (R genome) and a natural apomictic tetraploid accession of B. brizantha (B genome). Chromosomes associated predominantly as bivalents. From this phase to the end of meiosis, chromosomes presented irregular segregation and abnormal arrangement in the metaphase plate. During metaphase I, in 27.8% of meiocytes, bivalents were distributed in two metaphase plates. In anaphase I, two distinct and typical bipolar spindles were formed. In 29.7% of pollen mother cells, one genome did not divide synchronically, with chromosomes lagging behind or not segregating at all. The second division was very irregular, resulting in polyads. Based on previous results from analysis of a triploid hybrid between these species, where the R genome was eliminated by asynchrony during meiosis, it is suggested that the laggard genome in this hybrid also belongs to B. ruziziensis.

Chromosome number and meiotic behavior were evaluated in 36 Brazilian accessions of the grass Paspalum (which had never previously been analyzed) to determinate which accessions might be useful in interspecific hybridizations. The analysis showed that one accession of Paspalum coryphaeum was diploid (2n = 2x = 20) and one accession of Paspalum conspersum hexaploid (2n = 6x = 60), the remaining 34 accessions being tetraploid (2n = 4x = 40). The pairing configuration was typical for the ploidy level i.e. in the diploid, chromosomes paired as 10 bivalents, in tetraploids as bi-, tri- and quadrivalents, and in hexaploid as 30 bivalents. A low frequency of meiotic abnormalities (less than 10%) was observed in the diploid, hexaploid and some tetraploid accessions, although the majority of tetraploid accessions showed a high frequency of meiotic irregularities. The use of accessions with a low frequency of meiotic abnormalities in breeding programs is discussed.

Cytogenetic studies were carried out on 22 accessions of Brachiaria jubata from the Embrapa Beef Cattle Brachiaria collection. One accession was diploid (2n = 2x = 18) and the remaining 21 were tetraploid (2n = 4x = 36). Among five tetraploid accessions, a specific and constant pattern of cell fusion involving only two microsporocytes was recorded. Meiosis proceeded normally from prophase I to the end, giving rise to an octad with normal microspores that developed into fertile pollen grains. Regular octad formation was possible because each cellular chromosome set was maintained in its proper domain, spindles were correctly positioned, and cytokinesis planes were formed in the correct places. Such behavior of meiosis in syncytes has never been reported in any other plant species.

A meiose é um evento de alta estabilidade evolucionária que culmina na redução do número de cromossomos. O curso normal e harmonioso da meiose garante a viabilidade gamética. Os eventos citológicos da gametogênese são controlados por um grande número de genes que atuam desde as mitoses pré-meióticas até as pós-meióticas. Mutações nestes genes causam anomalias que podem afetar a fertilidade. Durante a avaliação do comportamento meiótico em algumas espécies de plantas, muitas anormalidades que afetaram a fertilidade ou causaram total macho-esterilidade foram detectadas. Algumas das anormalidades encontradas já eram descritas na literatura, enquanto outras eram totalmente desconhecidas. As anormalidades mais freqüentes nas espécies analisadas foram segregação irregular de cromossomos, citomixia, aderências cromossômicas, mixoploidia, fragmentação cromossômica, formação de sincícios, fusos anormais e falta de citocinese. Eliminação de cromossomos, uma anormalidade meiótica rara, foi observada em uma espécie de Paspalum. Mutantes meióticos nunca antes descritos, afetando diferentes passos da meiose, foram observados em algumas espécies, especialmente em milho, soja e Paspalum. Alguns mutantes apresentam características que podem ser exploradas com sucesso em programas de melhoramento, pois causam total macho-esterilidade.

Meiosis is an event of high evolutionary stability which culminates in a reduction of chromosome number. The normal and harmonious course of meiosis ensures gamete viability. The cytologic events of gametogenesis are controlled by a large number of genes that act from premeiotic to postmeiotic mitosis. Mutations in these genes cause anomalies that may impair fertility, and many abnormalities affecting plant fertility or causing total male sterility have been detected during the evaluation of meiotic behavior in some species. Some of these abnormalities have been frequently described in the literature, while others have not been previously reported. The most frequent abnormalities found in the species analyzed were irregular chromosome segregation, cytomixis, chromosome stickiness, mixoploidy, chromosome fragmentation, syncyte formation, abnormal spindles, and failure of cytokinesis. Uncommon abnormalities, such as chromosome elimination during microsporogenesis, were found in one species. Original meiotic mutations affecting different steps of meiosis were also observed in these species, especially in maize, Paspalum and soybean. Some mutants present characteristics that may be exploited successfully in breeding programs because they cause total male sterility.

Eliminação de cromossomos em híbridos interespecíficos é uma ferramenta poderosa em programas de melhoramento. Entre 12 variedades brasileiras de aveia recomendadas para cultivo na região Sul, uma (UFRGS 15) apresentou um tipo de eliminação cromossômica nunca antes descrita em outra espécie. Cromossomos que deram origem a micronúcleos durante a meiose foram eliminados do micrósporo como micrócito. O micronúcleo aproximava-se da parede do micrósporo, formava um tipo de brotamento e sofria separação do micrósporo. Os micrócitos eliminados deram origem a grãos de pólen pequenos e estéreis. As possíveis aplicações deste achado para o melhoramento de aveia são discutidas.

Chromosome elimination in interspecific hybrids is a powerful tool in breeding programs. Of 12 Brazilian oat varieties recommended for cultivation in southern Brazil, one variety, UFRGS 15, presented a new type of chromosome elimination never described in any other species. Chromosomes that produced micronuclei during meiosis were eliminated from microspores as microcytes. The micronucleus reached the microspore wall and formed a kind of bud, separated from the microspore. The eliminated microcytes gave origin to small and sterile pollen grains. The possible applications of this finding for oat breeding are discussed.

Quinze variedades de soja adaptadas ao cultivo para duas regiões do país foram citogeneticamente analisadas para melhor compreender a citologia desta cultura e aprimorar a técnica para estudos meióticos. A análise mostrou uma baixa freqüência de anormalidades meióticas e as variedades não foram igualmente afetadas. Segregação cromossômica irregular, aderências cromossômicas, conexões citoplasmáticas entre células, citomixia e fusos irregulares foram as anormalidades observadas entre quatorze variedades, nenhuma das quais foi descrita previamente em soja. Todas as anormalidades observadas podem afetar a fertilidade do pólen. A fertilidade do pólen foi alta e, exceto para algumas variedades, esteve correlacionada com as anormalidades meióticas. Embora soja não seja um sistema modelo para estudos citológicos, nosso primeiro contato com esta espécie mostrou que é possível a condução de estudos citogenéticos. Algumas modificações no método padrão para estudos meióticos foram necessárias para se obterem resultados satisfatórios.

Despite the importance of soybeans little cytogenetic work has traditionally been done, due to the small size and apparent similarity of the chromosomes. Fifteen soybean [Glycine max (L.) Merrill] varieties adapted for cultivation in two distinct regions of Brazil were analyzed cytogenetically. A low frequency of meiotic abnormalities was noted in all varieties, although they were not equally affected. Irregular chromosome segregation, chromosome stickiness, cytoplasmic connections between cells, cytomixis and irregular spindles were the main abnormalities observed, none of which had been described previously in soybeans. All of these abnormalities can affect pollen fertility. Pollen fertility was high in most varieties and was correlated with meiotic abnormalities. Although soybean is not a model system for cytological studies, we found that it is possible to conduct cytogenetic studies on this species, though some modifications in the standard methods for meiotic studies were necessary to obtain satisfactory results.

Apesar da importância econômica do gênero Paspalum, pouca ou nenhuma informação citológica é encontrada para a maioria das espécies. Esta é a primeira descrição sobre número de cromossomos e comportamento meiótico para P. maritimum. Os três acessos coletados no Estado do Amapá mostraram-se tetraplóides (2n=4x=40) com os cromossomos associando-se predominantemente como bivalentes. A baixa ocorrência de associações multivalentes sugere que estes acessos sejam alotetraplóides segmentais. Todos os acessos mostraram uma baixa frequência de anormalidades meióticas e, como consequência, uma alta fertilidade de pólen, mostrando potencial para serem utilizados em programas de hibridização.

Despite of economic importance of the genus Paspalum, little or no cytologic information is available for many species. This is the first report about chromosome number and meiotic behavior for P. maritimum. The three accessions collected in Amapá State (North Region of Brazil) were tetraploid (2n=4x=40) with the chromosomes associating predominantly as bivalents. The low frequency of multivalents suggested that they were segmental allotetraploids. All accessions showed a low rate of meiotic irregularities, and as a consequence the pollen fertility was high. The results suggested that these accessions presented potential for use in a hybridization program.

Com o objetivo de avaliar o efeito do stress ambiental causado pelo alto teor de alumínio encontrado nos solos de cerrado sobre o comportamento meiótico, um elevado número de genótipos de milho está sendo avaliado. Neste estudo relata-se a ocorrência de fusão celular, seguida ou não de degeneração cromatínica, em meiócitos de algumas plantas de uma linhagem endogâmica cultivada em solo ácido e corrigido no planalto central Brasileiro. Os resultados não sugerem qualquer efeito do solo sobre as irregularidades observadas, pois o número de plantas afetadas foi maior dentre aquelas cultivadas em solo corrigido. As possíveis causas destas irregularidades e suas influências na produção de macho-esterilidade são discutidas.

A large number of maize genotypes are currently being evaluated to determine the effect of environmental stress caused by the aluminum found in cerrado soils on meiotic behavior. In the present study we report the occurrence of cell fusion followed or not by chromatin degeneration in meiocytes of some plants of an inbred line of maize grown on acid and corrected soil in the Brazilian central plateau. The results suggest the absence of any soil effect on the irregularities observed since the number of affected plants was higher among those grown on corrected soil. The possible causes of these irregularities and their influence on male sterility are discussed.
